Well the truck got a code for a misfire in cylinder 2 is cylinder 2 located at the front of the engine corner. the trucks not mine helping my grandpa. Diagnose the problem going to check the wire harness and spark plug along with vacuum leaks around the manifold and maybe even check for compression in that cylinder. What else should i be looking for?

A misfire can be due to weak/lack of spark, fuel or compression. I would start with a compression test in that cylinder. As you remove the plug for the test check it’s condition. Check the wire as well. Then do the compression test. If that comes back good then I would look to the ignition coil. Here, swap the coil from cylinder 2 to another cylinder. If the coil is defective, the misfire should move to the defective cylinder. Now, if the coil is fine check out the injector on 2. It is a single cylinder misfire so that narrows a potential fuel problem to the injector. Check for leaking injector seals etc.
